A karyotype analysis has been done using squash-acetoorcein method. Shape and size of prometaphase chromosomes were successfully determined The chromosomes length of salak were 1.15 pm � 2.38 pm. The suggested karyotype formula of salak was 2n = 28 = 11 m + 1 m (SAT) + 2 sm, which meant the disomic state was eleven pairs of metacentric chromosomes, single pair of metacentric chromosomes bearing satellite, and two pairs of submetacentric chromosomes. The intrachromosomal assymetry index was 0.3 f 0.03, and the interchromosomal assymetry index was 0.2 ± 0.02. Some chromosome pairs had similar size and shape that were difficult to distinguish. Further identification using chromosomal banding is necessary If:Pills* the result. Key words: salak [Salacca zalacca (Gaertner) Voss.], chromosome, karyotype analysis. INTISARI Penelitian sitogenetika tanaman salak (Salacca zalacca {Gaertner] Voss) merupakan aspek penting dalam analisis genetika tanaman salak dan menunjang program pemuliaan untuk merakit kultivar salak unggul. Analisis kariotipe kromosom salak telah dilakukan dengan metode squash dan pewarnaan aceto-orcein. Dengan metode ini, pengamatan morfologi kromosom, yakni ukuran dan bentuk kromosom, dapat dilakukan dengan hasil baik pada stadia prometafase. Hasil perigamatan menunjukkan bahwa panjang kromosom salak 1,15 mm � 2,38 mm. l3erdasarkan hasil analisis sifat morfologi kromosom, rumus kariotipe salak adalah 2n = 28 = 11 m + I m (SAT) + 2 sm, yang berarti adalah sebelas pasang kromosom metasentrik, satu pasang kromosom metasentrik dengan satelit, dan dua pasang kromosom submetasentrik. Indeks asimetri kariotipe intrakromosom adalah 0,3 ± 0,03 dan indeks asimetri interkromosom adalah 0,2 ± 0,02. Beberapa pasangan kromosom salak mempunyai bentuk dan ukuran yang mirip sehingga sulit dibedakan. ldentifikasi kromosom tanaman salak dengan teknik pemitaan kromosom (chromosomal banding) perlu dilakukan untuk mendukung hasil yang telah dicapai.
