description Melanocortin 4 Receptor (MC4R) is one of candidates gene for growth traits and feed intake in mammals. The objective of this research was to identify the genotype of Menocortin 4 Receptor gene in Bligon goat. Blood samples from 40 Bligon goats were used for DNA extraction. Primer used was designed from Capra hircus genome in Genbank Accession Number NM_001285591. Primer CapraPF:5’-ATTTCCAAGTGATGCCGACC’3 and CapraPR:5’- CTCCAACAAGCTGATGACCC-‘3 were used to amplify 387 bp of PCR product. Using DNA sequencing, a Single Nucleotide Polymorphism (SNP) was detected in Bligon goat. The SNP identified in this research was g.256 G/A. The results of DNA sequence of PCR product in Bligon goats were used for restriction map of specific enzyme using Bioedit version 7.2.0. Based on restriction mapping, HpyCH4V (A'CG_T) was detected to recognize SNP region. Rectriction enzyme HpyCH4V can be usedfor genotyping of targeted gene using PCR-RFLP method and it will be assosiated with growth traits and feed intake in Bligon goat.
