Summary:The aim of this study was to find out the plasmid profile of the antibiotic resistant E. coli, to ampicillin, streptomycin and enrofloxacin. Eight of E. coli isolates have been identified and further assessed for sensitivity to these antibiotics, which then were isolated their plasmids.The antibiotics resitant E. coli were cultured on lactose broth, incubated in a shaker incubator at 37°Covernight. Cell collection has been done by centrifugation at 12.000rpm for 5 minutes two times. Plasmid isolation was carried out by lyses alkali method, using solution I, II, and III. Plasmid precipitation was done by added 00 MNa acetate and ethanol absolute. Plasmid profile was performed on 1% agarose on electrophoretic analysis using plasmid marker. The result indicated that the E. coli resistant antibiotics have one to three of plamids and it's profile appears as flourescent pita upon ultraviolet radiation, which indicated as a mega plasmid on the 4268 bp, 4973 bp, 5148 bp, and between 5148 bp -21226 bp. Each E. coli isolate showed 1to 3DNA pitas of plasmids.
