description Insulin is a hormone existence secreted by Langerhans pancreatic beta cells and plays an important role in the metabolism of carbohydrates, fats and proteins. The presence of insulin in the hippocampus may indicate its involvement in brain cognitive functions, such as learning and memory phenomena. The purpose of this study is to watch the density of insulin immunoreactive cells in pancreas and hippocampus formation. In addition, this study also observe the body weight and fasting blood glucose levels in Balb-C mice which treated with streptozocin repeated as the animal model of diabetes mellitus. Thirty male mice Balb-C strain, aged 12-14 weeks, weight 30-40 g, divided into 2 treatment groups, each group consisted of 15 individuals. K1 (control) were treated a sodium citrate buffer, while K2 (diabetes mellitus) is treated a streptozotocin at a dose of 40 mg/kg bw in 50 mM sodium citrate buffer pH 4.5 in intra-peritoneal of 0.5 ml for 5 consecutive days. The criteria of body weight and fasting blood glucose levels were examined on day 0, 7, 14, 21 and 28 after administration of treatment. Every two animals from each group of the experiment were done euthanasia on day 7, 14, 21 and 28 respectively after the administration of treatment, subsequently the mice were done a perfusion of fixative solution and a necropsy to take the pancreas and brain tissues for examination immunohistochemically using anti-insulin mouse antibody. Based on statistical analysis using ANOVA, the density of insulin immunoreactive cells in pancreatic and hippocampus formation of group 1 were higher than group 2 which showed a significant difference (p<0.05), as well as the time of observation and interaction between group and time showed significant differences (p<0.05). The levels of fasting blood glucose in group 2 increased than group 1 with significant difference (p<0.05). Decline of the body weight in group 2 differed significantly to group 1 (p<0.05). The levels fasting blood glucose and body weight at each time of observation and interaction between group and time of observation also showed significant differences (p<0.05).
