Dental caries cavity swab on the media's first permanent mandibular molars. Probiotic Lactobacillus reuteri Prodentis possessesin vivo antimicrobial activity against Streptococcus mutans present in the oral cavity and might be used asan alternative measure to prevent dental caries. This study is aimed to determine the effect of probiotic Lactobacillusreuter:iProdentis on the growth of Streptococcus mutans colony. This research uses laboratory experimental method with pre-post test control group design. The data obtained were analyzed using parametric statistical tests, namely t-group (paired sample t-test) to determine the differences in pre-post data on treatment. The results of this study said that the giving of probiotic Lactobacillus reuteri Prodentis gives significantdifferences in the decrease of the number of colonies of 1 week treatment are indicated by the value of p &lt
