Summary: | Black Orchid (Coelogyne pandurata Lindley) is an endemic orchid from
Kalimantan. It has highly commercial value because of the uniqueness of its black
labellum. Recently, it has been considered threaten to be extinct due to
overcollection and land use conversion. Mass propagation of this orchid using
conventional methods is still limited, so that a modern method is needed. For this
purpose, transgenic technology with a key gene for shoot induction can be
applied. The aim of this research is to improve the capability of shoot formation of
the Black Orchid through genetic transformation of KNAT1 gene by
Agrobacterium tumefaciens.
The research was conducted in 5 steps: 1. Decided the most responsive
developing protocorms and the best germination phase of protocorms as the
