Summary: | Agriculture products are susceptible contaminated with ochratoxin A (OTA),
commonly produced by several fungi such as Penicillium verrucosum, Penicillium
nordicum, Aspergillus section nigri or black aspergilli especially A.carbonarius,
dan A.niger. This research was focused on black aspergilli isolated from
fermented cocoa, coffee beans and dried cassava in Yogyakarta, and designed to
identify species within black aspergilli group and their OTA ability. Identification
black aspergilli at species level will be useful to make clear relationship between
OTA contamination on food product and the toxin producer. Identification of the
isolate were carried out by using macroscopic as well as microscopic characters
on identification media and confirmed by specific PCR assays. Four isolates
(YAC-9, YAK-6, YAK-12, YAG-3) were found as OTA producing-strain, with
the highest OTA found on YES solid medium was 57,68 ppb. Based on
morphological characters, 16 isolates can be grouped into 5 species, after
confirmed by molecular data, the groups was identified as A.carbonarius, A.
niger, A. tubingensis, A aculeatus, dan A.ibericus. OTA producing-strains were
identified as A.carbonarius and A. niger, meanwhile A. tubingensis, A aculeatus,
dan A.ibericus was found non OTA producing-strains.
