Ethyl acetate extract of fresh citrus fruit peel is known to have a greater antibacterial activity than essential oil. Therefore, studies of antibacterial activity of ethyl acetate extract of Citrus spp. fruit peel and ethyl acetate fraction rest of the most active extract distillation against acne-inducing bacteria and identification of active compound was done. Antibacterial activity of ethyl acetate extract of Citrus aurantifolia (Christm. & Panz.) Swingle (lime), Citrus hystrix DC. (kaffir lime), and Citrus nobilis Lour. (tangerines) fresh fruit peel were tested against Propionibacterium acnes ATCC 6919, Staphylococcus aureus ATCC 25923, and Staphylococcus epidermidis by pour plate diffusion method. The most active extract (kaffir lime rind) was distilled by water distillation to separate essential oil fraction, then distillation residue was partitioned with ethyl acetate to obtain ethyl acetate and water fractions. MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) values of the most active fraction (ethyl acetate fraction) were determined by broth dilution method. Spot of ethyl acetate fraction that has antibacterial proved by bioautography test was isolated using preparative TLC (thin layer chromatography). Structure of active isolate was identified based on UV, IR, 1H NMR, 13C NMR, 1H-13C HMQC, 1H-1H COSY, 1H- 13C HMBC, MS spectra and compared to data from literature. MIC and MBC values of ethyl acetate fraction against S. epidermidis were 1.13 mg/ml and 2.25 mg/ml, respectively, while MIC and MBC values of ethyl acetate fraction against S. aureus were 0.56 mg/ml and 1.13 mg/ml, respectively. Chemical structure of active isolate was identified as furanocoumarin oxypeucedanin hydrate or 5-(2,3-dihydroxy-3-methylbutoxy) psoralen.
