description This research is a true designed experiment, aimed to develop a new (novel) method in preparation of fresh sperms of goat that able to maintain the functional viability and fertility of spermatozoa at storage temperature of 14 15°C for 14 days, and can be able to applied for AI effectively and efficientlly. The study is targeted as a solution for the failure of the implementation of AI in the area that is the presence of frozen sperm and liquid nitrogen are limited. The research was carried out in three stages, in which the fresh sperm preparation method through supplementation of various doses of Leucocepahala tannin (LT) per ml of semen with extender glucose 18.9%, as phase I, the replacement of seminal plasma with Earle'S medium supplement (EarleS balanced salt solution: EBSS) and 100 μl hypotaurine-2mM l(-1) EBSS, as phase II, and the AI applications using fresh sperm from experiment I and II, as phase III. The results showed that supplementation of LT2,5% per ml of semen in the extender 18.9% glucose and seminal plasma replacement with EBSS medium, a fresh sperm of goat preparation method is best in terms of recovery on the percentage of sperm viability and normal morphology on the storage. Both method improve pregnancy rate (conception rate: CR) resulteds from the first AI service of 95.38% and 100%, and the kidding rate (KR) are 75.38% and 72.72%, respectively.
