Summary: | Background: Diffuse Large B cell Lymphoma (DLBCL) is the most common
malignant tumor of lymphoid tissue derived from the mature B cells. Based on the
location, DLBCL can be divided into nodal and extranodal DLBCL, and they
proposed differently to have the clinical behaviors and prognostic. They are
caused by many factors including because of the proliferation and apoptosis of
tumor cells. The markers of proliferation and apoptosis are used the proteins of
P53 and KI-67, and the expressions of P53 and KI-67 can be used to distinguish
the aggressiveness of nodal and extranodal DLBCL. Various studies on the
expressions of P53 and KI-67 DLBCL showed controversial results.
Objectives: To investigate the difference mean of P53 and Ki67 protein
expressions between nodal and extranodal DLBCL.
Methods: This study was a retrospective cross sectional. The total sample size of
40 DLBCL samples were taken from Sardjitos Hospital at Yogyakarta that
consist of 21 nodal DLBCL and 19 extranodal DLBCL samples respectively. The
preparation was stained using immunohistochemistry with monoclonal P53 and
KI-67antibody.
The expression of P53 and Ki-67 was calculated from the number of positive cell
nuclei are stained brown. The differences mean of the P53 and Ki67 DLBCL of
nodal and extranodal were analyzed using Independent Sample t-Test.
Results: In this study find no significant difference between age clinical
characteristics and gender to nodal and extranodal locations (p = 0.527, p =
0.1000). This study also find not significant differences mean in P53 expression to
nodal and extranodal DLBCL (p = 0.778), but the mean of Ki-67 expression to
nodal and extranodal DLBCL significant difference (p = 0.04).
Conclusion: The study find not significant differences between nodal and
extranodal DLBCL in terms of age, gender and P53 expression, but they
significant difference in Ki67 expression. The mean of Ki-67 nodal expression is
higher than the mean of Ki-67extranodal expression which indicate that nodal
DLBCL more aggressive than extranodal DLBCL.
