Designing inhibitors of M2 proton channel against H1N1 swine influenza virus. by Qi-Shi Du, Ri-Bo Huang, Shu-Qing Wang, Kuo-Chen Chou

    Published 2010-02-01
“…The computer-aided drug design methods are used to calculate the binding free energies, with the computational biology techniques to analyze the interactions between M2 proton channel and adamantine-based inhibitors.1) The NMR structure of M2 proton channel provides a reliable structural basis for rational drug design against influenza virus. 2) The channel gating mechanism and the inhibiting mechanism of M2 proton channel, revealed by the NMR structure of M2 proton channel, provides the new ideas for channel inhibitor design. 3) The newly designed adamantane-based inhibitors based on the modeled structure of H1N1-M2 proton channel have two pharmacophore groups, which act like a "barrel hoop", holding two adjacent helices of the H1N1-M2 tetramer through the two pharmacophore groups outside the channel. 4) The inhibitors with such binding mechanism may overcome the drug resistance problem of influenza A virus to the adamantane-based drugs.…”

    Published 2020-12-01
“…The Current River in Missouri (USA) contains a single anthropogenic barrier to longitudinal connectivity: the Cedar Grove low-water crossing, which spans the main channel (10 culverts) and a side channel (4 culverts). In July 2017, we radio-tagged Northern Hog Sucker Hypentelium nigricans upstream (henceforth ‘above’; N = 24) and downstream (henceforth ‘below’; N = 26) of the crossing and followed their movements monthly for a year to assess fish passage and maximum displacement. …”

Characterizing the role of atrial natriuretic peptide signaling in the development of embryonic ventricular conduction system by Arun Govindapillai, Adam Hotchkiss, Mark Baguma-Nibasheka, Robert A. Rose, Lucile Miquerol, Oliver Smithies, Nobuyo Maeda, Kishore B. S. Pasumarthi

    Published 2018-05-01
“…The present study showed that treatment of embryonic ventricular cells with ANP or cell permeable 8-Br-cGMP can induce gene expression of important VCS markers such as hyperpolarization-activated cyclic nucleotide-gated channel-4 (HCN4) and connexin 40 (Cx40). Inhibition of protein kinase G (PKG) via Rp-8-pCPT-cGMPS further confirmed the role of ANP/NPRA/cGMP/PKG pathway in the regulation of HCN4 and Cx40 gene expression. …”
