Preparation of enteroinvasive Escherichia coli reference materials for food analysis
Published 2021-11-01“…ObjectiveTo satisfy the demands of laboratory quality control and proficiency testing for the detection of enteroinvasive Escherichia coli (EIEC), a stable EIEC reference material was developed with independent intellectual property rights, good stability and clear background genomic information.MethodsNGS Technique was used to sequence the whole genome of enteroinvasive Escherichia coli (CMCC 44840), bioinformatics analysis was performed to identify the species, serotype, MLST type and virulence genes; EIEC lyophilized samples with 103 CFU were prepared by freeze-drying method ; The homogeneity test was performed according to CNAS-GL017, and the result were statistically analyzed by one-way ANOVA; The samples were stored at -20, 4, 25 and 37 ℃, respectively, and their storage stability and transport stability were evaluated; Five kinds of food matrix samples were used to verify the applicability of reference materials, and three laboratories were organized for collaborative calibration.ResultsThe genome size of CMCC 44840 was 4.96 Mb, the GC content was 50.7%, and the genome contained 5424 coding genes. …”

Quantification of Minimal Disease by Digital PCR in ALK-Positive Anaplastic Large Cell Lymphoma: A Step towards Risk Stratification in International Trials?
Published 2022-03-01“…We aimed to assess droplet digital PCR for quantification of minimal disease in an inter-laboratory setting in a large cohort of 208 uniformly treated ALCL patients. Inter-laboratory quality control showed high concordance. Using a previously described cut-off of 30 copies <i>NPM-ALK/</i>104 copies <i>ABL1</i> (NCN) in bone marrow and peripheral blood, MDD quantification allowed identification of very high-risk patients (5-year PFS% 34 ± 5 for patients with ≥30 NCN compared to 74 ± 6 and 76 ± 5 for patients with negative or <30 NCN, respectively, <i>p</i> < 0.0001). …”
