This experiment was conducted to determine the rumen fermentation parameters (pH, NH3 and VFA) and rumen microbial protein synthesis of Ongole Grade cattle and Buffalo. Three female rumen fistulated Ongole Grade cattle and Buffalo were used for investigating rumen fermentation parameters and three female Ongole Grade cattle and buffalo for estimating rumen microbial protein synthesis. All of them were treated by single feed of glyricidia (G), corn straw (CS) and calyandra (C) ad libitum. The result, showed that the NH3 consentration for Buffalo (17,01 mg/100 ml) was higher (P< 0,01) than Ongole Grade cattle (8,91 mg/100 ml), and NH3 consentration for glyricidia (22,31 mg/100 ml) was higher (P< 0,01) than calyandra (10,17 mg/100 ml) and corn straw (6,41 mg/100 ml). Consentration of VFA for Buffalo (82,32 mmol/1) was higher (P< 0,01) than Ongole Grade cattle (55,81 mmol/1), but consentration of VFA were not significantly different among feed. Microbial protein synthesis for Ongole Grade cattle (22,92 g N/kg RDOM*) was higher than Buffalo (8,41 g N/kg RDOM), however microbial protein synthesis for glyrisidia (25,12 g N/kg RDOM) was higher (P< 0,01) than calyandra (13,97 g N/kg RDOM) and corn straw (7,12 g N/kg RDOM)
